The day before the test, transfer the bacteria, which have been subcultured on a slant medium, to liquid SCD medium (10 mL test tube) using 1 oz. culture, and pre-culture at 30-35°C for 18-24 hours. Then, serially dilute with peptone-sodium chloride buffer to adjust to the appropriate concentration before use in the test.
